Components:

Phloxine B Solution 0.5%

Methylene Blue Azure B Solution

Acetic Acid 0.2% Aqueous


Fixation:

Any well fixed tissue.


Section:

Paraffin at 6µm


Staining:

  1.  De-paraffinised and hydrate to distilled water.                          
  2. Stain for two minutes in Phloxine B Solution, 0.5%. Rinse well in distilled water.
  3. Stain for 1 minutes in Methylene Blue-Azure B Solution.
  4. Differentiate in Acetic Acid 0.2%.
  5. Complete the differentiation in three changes of 95% alcohol.
  6. Dehydrate in absolute alcohol and clear in xylene, two changes each.
  7. Mount with Permount.

Plasma Cell Cytoplasm

Blue

Malarial Parasites

Blue

Erythrocytes

Pink

Other Tissue Elements

Shades of rose to red
